In vivo tests for a carcinogenic metabolite in plasma of animals given injections of ethyl carbamate (urethan).

چکیده
Plasma, obtained from rabbits given injections of urethan 2J hours previously, when administered intraperitoneally to Swiss mice, produced a significantly increased inci dence of lung adenomas in these animals, as well as an increased incidence of skin papillomas at the site of subsequent croton oil applications. These effects were not obtained when the plasma was lyophilized or desiccated, with the consequent removal of the free urethan. The condensed vapors collected dur ing the process of lyophilization, on the other hand, were carcinogenic. Carbonyl-labeled urethan-C14 was used to determine the urethan content of the

Comparative carcinogenicities and mutagenicities of vinyl carbamate, ethyl carbamate, and ethyl N-hydroxycarbamate.
Structural modifications of the multipotential carcinogen ethyl carbamate (urethan), CH3CH2—O---CO—-NH2, generally result in dramatic losses of carcinogenic activity, which usually have been assayed by lung adenoma induction and by initiation of skin papillomas in mice (reviewed in Refs. 12 and 20). One exception is ethyl N-hydroxycanbamate, CH3CH2— O—CO—-NHOH, a metabolite of ethyl c...

Ethyl carbamate and a series of related compounds were given in single intraperitoneal doses to pregnant Syrian hamsters on Day 8 of gestation, and fetuses were examined for malformations on Day 13 of gestation. Urethan produced a variety of malforma tions and growth retardation. Of the compounds modified in the carboethoxy end, n-propyl carbamate was as teratogenic as the ethyl carbamate, and ...

متن کاملVinyl carbamate as a promutagen and a more carcinogenic analog of ethyl carbamate.
Vinyl carbamate was much more active (10 to 50 times) than ethyl carbamate for the initiation of skin tumors and for the induction of lung adenomas in mice. Vinyl carbamate was also mutagenic to Salmonella typhimurium TA 1535 and TA 100 in the presence of reduced nicotinamide adenine dinucleotide phosphate-fortified rat or mouse liver mitochondrial supernatant fractions. This mutagenic activity...
متن کاملEffects of ethyl carbamate (urethan) on the early development of the teleost Brachydanio rerio.
INTRODUCTION The narcotic urethan (ethyl carbamate) has proved of increasing interest to the cellular physiologist since the first demonstration of its carcinogenic properties (29) in 1943 and the subse quent announcement of its carcinoclastic activities (16) in 1946. It has been shown to arrest or reduce mitotic activity in the eggs of sea urchins (9, 13, 25) and in ciliates (7, 31), as well a...
